Here is a comparison (2 sisters are being compared).
Comparison always uses comparative form so it'll be 'of the two sisters Meena is elder'.
'Of the two sisters Meena is the eldest' will also be wrong because here is comparison and comparison never takes superlative degree.

How is possible?
There is a rule in adjective if two is given the comparative & superlative degree is used.
Eg:Sita is the faster of the two sisters.
