I dont think the second box is correctly labelled. If you swivle the side that holds a '3' to the front and with a '6' on the top, the box that shows a '2' in the diagram should actually be a '5'... shouldn't it?

By combining box 3 and 4, I think the number opposite to 3 should be 2.

That's correct. The opposite number to 3 means 5 only.
