I think option A. Sentence itself showing the superlative form of population of Tokyo. Using among, city's population can be easily compared.

Because here we are comparing Tokyo with all other towns in the world. So we describe with among over here.

Ya its so simple, in this sentence they are comparing population of Tokyo with other countries. And this is the Grammar rule If you are comparing the same thing of other country so we shall use than that of because they are comparing the population with population.

"that of" in the sentence has no significance as such so the and should be D.

In the case of 2 countries greater should be used but for more country greatest should be used. So the answer may be option A.
