Over is wrong because you can't smile over something in this instance fate. So over is wrong and and on will be the right answers. Fate is something that is going to come or already happened so over is wrong.

Smile is a verb(a word defining an action). Actions can take all of the given choices (prepositions).
But here the context is such that, the subject is Fate (a bigger entity as we see it). Therefore, fate can only smile 'at' us or smile 'on' or 'upon' us (all positive sentences).
It won't smile with us (as it's bigger than us) and never smile over us (as this makes the sentence negative ).

Most of you are getting it wrong. This sentence [if I am correct] is a saying. This sentence means 'fate favours'. The smile is not seen-it does not have to be physical.
