"Smile on" for this sentence would be incorrect as it gives a negative effect. It means that u r trying to tease someone or making fun of someone. Here the sentence says that fate will favor those who have urge to complete their work/duty irrespective of all hurdles.
Ex:
Smile on you -- incorrect.
Smile with you. correct.
Open with smile - correct.
Open on smile - incorrect.
Smile on your face - correct.
Smile with your face - incorrect.
It simply depends upon the situation.

Smile is a verb(a word defining an action). Actions can take all of the given choices (prepositions).
But here the context is such that, the subject is Fate (a bigger entity as we see it). Therefore, fate can only smile 'at' us or smile 'on' or 'upon' us (all positive sentences).
It won't smile with us (as it's bigger than us) and never smile over us (as this makes the sentence negative ).

I let you here why "on a preposition is used" in English, the "preposition the 'on' is commonly used to denote support, favor, or attention directed towards someone or something. Therefore in this sentence 'on' is used to convey that fate's favourable disposition (tendency) is directed toward those who face life's harsh realities and perseverance".

Over is wrong because you can't smile over something in this instance fate. So over is wrong and and on will be the right answers. Fate is something that is going to come or already happened so over is wrong.

Most of you are getting it wrong. This sentence [if I am correct] is a saying. This sentence means 'fate favours'. The smile is not seen-it does not have to be physical.

"Smiles with" looks best as "smiles on" and "smiles over" suggest as if you're making fun of someone and that is not the idea here.

I think on is right as smile is an expression and make it is the best preposition which can be used in that situation.
