"Smile on" for this sentence would be incorrect as it gives a negative effect. It means that u r trying to tease someone or making fun of someone. Here the sentence says that fate will favor those who have urge to complete their work/duty irrespective of all hurdles.

Ex:

Smile on you -- incorrect.
Smile with you. correct.

Open with smile - correct.
Open on smile - incorrect.

Smile on your face - correct.
Smile with your face - incorrect.

It simply depends upon the situation.

Most of you are getting it wrong. This sentence [if I am correct] is a saying. This sentence means 'fate favours'. The smile is not seen-it does not have to be physical.
