Mutation can only be studied first by looking t the expression of the gene ( here phenotype )so it reflects the mutation but when you study the single nucleotide poly morphism (snps) by Restriction Enzyme experiment you will realize where the mutation is.

Some time phenotype may change beside genotype does not, this is studied in 'epigenetics'. Henceforth, mutation can be defined with the C option suitably.
