An Enterprise is a large business, school, government, etc. Entity. An Enterprise system is one that is geared towards heavy use, with appropriate licensing and performance, that generally meets or exceeds standards (such as ANSI SQL, XML, etc) for that kind of system.
MS SQL Server, Oracle, IBM DB2, are all enterprise databases. MS Access is generally not considered one, it is more of a single or limited user system.

Database applications cannot access database data directly. They interacts with the DBMS which in turn access data from the database with some query language.
